The CD1d protein complex, a non-classical class I MHC found on various immune cells, facilitates the presentation of lipid antigens to natural killer T cells (NKT). In particular, invariant natural killer T (iNKT) cells, a subset of NKT cells, express a semi-invariant T-cell receptor (TCR) and are distinguished by their reactivity to the lipid α-Galactosylceramide (α-GalCer) presented by CD1d.
iNKT cells play a crucial role in modulating immune responses to infections, tumors, and autoimmunity. Their potential use in cancer cell therapy is being explored due to their non-alloreactive nature and ability to infiltrate solid tumor tissue, coordinating anti-tumor activity with various immune cells.
CD1d Dextramer® is made up of a flexible dextran polymer decorated with multiple fluorophores and an optimized number of CD1d molecules. When loaded with α-Galactosylceramide (α-GalCer) the resulting dextramer is a highly sensitive reagent for detection and enumeration iNKT cells by flow cytometry
CD1d Dextramer® is also available unloaded, without a lipid, which can be used as a negative control for your experiment.
